ROS Detection Assay Kit (DCFDA / H2DCFDA)

The ROS Detection Assay Kit is a highly sensitive and safe assay to measure Reactive Oxygen Species (ROS). It employs the cell-permeant reagent Dichlorodihydrofluorescein-diacetate (H2DCFDA), a fluorogenic dye designed to measure the activity of hydroxyl, peroxyl, and other reactive oxygen species within cells. Once H2DCFDA diffuses into the cell, intracellular esterases cleave the acetyl groups, rendering it a non-fluorescent compound. This compound is then rapidly oxidized by ROS to a highly fluorescent 2′,7′-Dichlorodihydrofluorescein. The resulting fluorescence intensity is directly proportional to the ROS levels within the cell cytosol. The kit offers a fast, easy, and non-radioactive protocol.

Key Features:

  • Mechanism: Based on the unique fluorescent probe, H2DCFDA.
  • Protocol: Simple and fast, ensuring a user-friendly experience.
  • Safety: Non-radioactive, requiring no special handling.
  • Results: Provides high-throughput, accurate, and reproducible results.
  • Storage: Store at 4°C upon receipt.

Established Applications:

  1. Oxidative Stress Research: The kit is widely used to study cellular conditions related to oxidative stress, including apoptosis and aging.
  2. Disease Research: It is a valuable tool for investigating disease pathways, such as those involved in cancer, diabetes, and atherosclerosis, where ROS activity is elevated.
  3. Flow Cytometry: The H2DCFDA probe can be used in flow cytometry to analyze ROS levels in a population of cells, enabling high-throughput analysis.
