Capping B Solution – Tetrahydrofuran : Pyridine : N-Methylimidazole (80:10:10 v/v/v)
This Capping Reagent B is a three-component solution composed of tetrahydrofuran (THF), pyridine, and N-methylimidazole, mixed in an 80:10:10 volume ratio. It is used in the capping step of solid-phase oligonucleotide synthesis to activate Capping Reagent A (acetic anhydride), enabling acetylation of unreacted 5′-hydroxyl groups.
-
Composition: THF : Pyridine : N-methylimidazole (80:10:10 v/v/v)
-
Function: Activates Capping Reagent A for effective 5’-OH capping
-
Application: Capping solution for DNA/RNA synthesis workflows
-
Instrument Compatibility: Suitable for most automated synthesizers
-
Formulation: Ready-to-use
This formulation provides a more solvent-rich, low-viscosity matrix compared to traditional acetonitrile-based capping reagents, which may be advantageous for certain synthesis protocols or platforms.
