This assay operates on a simple yet effective principle: β-galactosidase catalyzes the hydrolysis of orthonitrophenyl-β-D-galactopyranoside (ONPG) into ortho-nitrophenol (ONP). This reaction produces a distinct bright yellow color, which can be detected and measured spectrophotometrically at 420 nm. The intensity of the resulting color is directly proportional to the concentration of β-galactosidase present in your sample, ensuring reliable and quantitative results.
