The PickMutant™ Random Mutagenesis Kit from Canvax provides a reliable, robust, and highly efficient method for introducing random mutations into DNA. This kit addresses the limitations of standard PCR polymerases, which typically produce insufficient mutation rates for generating diverse mutant libraries crucial for research.

Mechanism of Action:

This kit is based on the innovative incorporation of the mutagenic nucleotide analog dPTP into an amplified DNA fragment during PCR. A subsequent second PCR step, performed solely with natural dNTPs, effectively eliminates the dPTPs, leading to an impressive mutagenesis rate of up to 20%. dPTP serves as an excellent substrate for Taq Polymerase, preferentially incorporating in place of TTP and, with lower efficiency, in place of dCTP. This mechanism yields four types of transitions (A→G, T→C, G→A, and C→T), with A→G and T→C transitions occurring at higher frequencies, enabling comprehensive library generation.
